Lipidomic dataset of optic nerves from the silicone oil-induced ocular hypertension mouse model.

Glaucoma is characterized by progressive degeneration of retinal ganglion cells and their optic-nerve axons, with elevated intraocular pressure (IOP) as a major risk factor for retinal ganglion cell death. Lipids are essential to axonal structure and signalling, yet optic nerve lipidomic datasets under high IOP are limited. We present a stage-specific lipidomic dataset from a mouse model of silicone oil (SO)-induced ocular hypertension (high IOP) with contralateral balanced salt solution (BSS)-injected controls. Optic nerves from high-IOP eyes and matched contralateral controls were harvested at 2 weeks and 8 weeks post-injection to represent early and established optic nerve degeneration, with n = 6 per time point (3 males, 3 females). Lipids were extracted by a modified Bligh Dyer protocol and profiled by untargeted LC-MS/MS on a Vanquish Horizon ultrahigh performance liquid chromatography (UHPLC) coupled to a Thermo Q Exactive Orbitrap. Raw files were processed in LipidSearch 5.1, and statistical analysis was conducted through MetaboAnalyst 6.0. This data is available through Metabolomics Workbench, Study ID: ST004446.
