Refractory wounds impose a significant burden on patients and society. Although diagnostic ultrasound combined with microbubble contrast agents (USMB) improves microcirculation, its role in wound healing-particularly the underlying mechanisms-remains unclear. Here, we established a rat model of acute wounds and demonstrated that USMB therapy accelerated re-epithelialization and collagen deposition, accompanied by increased CD31 and VEGF expression, reduced IL-1β and TNF-α, and elevated TGF-β. By analyzing miRNA profiles, we revealed that USMB treatment was associated with the downregulation of miR-26b-5p. This molecule may target Gsk3β, thereby affecting the PI3K/Akt/GSK-3β signaling pathway and participating in USMB-mediated wound repair. These findings clarify the potential of USMB in wound therapy.
