Abstract
Here, we present a protocol for studying liver regeneration in adult zebrafish using a hepatic cryoinjury technique. We detail the surgical procedure to induce localized necrosis using a cryoprobe. We then detail steps for liver harvesting and CUBIC-based tissue clearing for three-dimensional imaging. The protocol enables comparison of healthy and injured liver parenchyma within individual animals, recapitulating key features of human chronic liver disease, including fibrosis formation and resolution.
