WB	Western blot analysis of extracts from C6 (lane 2(20μg), C6 (lane 3(40μg), using TriMethyl-Histone H3 Antibody. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody and Loading control antibody (Beta Actin, HY-P80438, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.
ICC	Immunocytochemistry analysis of Hela cells labeling TriMethyl-Histone H3 (Lys27) Antibody ( HY-P80922 ) at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with TriMethyl-Histone H3 (Lys27) Antibody ( HY-P80922 ) at 1/50 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L（HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue). 功能：三甲基化组蛋白 H3 是核小体的核心组分。核小体可将 DNA 缠绕并压缩为染色质，限制以 DNA 为模板的细胞机制对 DNA 的访问。因此组蛋白在转录调控、DNA 修复、DNA 复制和染色体稳定性中发挥核心作用。DNA 的可及性通过一系列复杂的组蛋白翻译后修饰 (也称为组蛋白密码) 以及核小体重塑来调控。亚细胞定位：细胞核；染色体亚基：核小体是由组蛋白八聚体组成，包含两个 H2A、两个 H2B、两个 H3 和两个 H4 分子，组装成一个 H3-H4 异源四聚体和两个 H2A-H2B 异源二聚体。该八聚体包裹约 147 bp 的 DNA。核小体与 TONSL、CHAF1A、CHAF1B、MCM2 和 DNAJC9 相互作用 (PubMed:33857403)。核小体还与 NASP 相互作用；NASP 是一种组蛋白伴侣，能够稳定并维持可溶性组蛋白 H3-H4 二聚体 (PubMed:22195965)。
