Abstract
OBJECTIVE: We aimed to investigate the association of the triglyceride-glucose index (TyG Index) and the single nucleotide polymorphism (SNP) of ALOX12, rs112667 with Type 2 Diabetes(T2DM) and Diabetic Kidney Disease(DKD) among individuals living in Shanghai, China. MATERIAL AND METHODS: This is a case-control study. In total, 200 subjects with T2DM who underwent physical examination in Jiuting Hospital from July 2021 to September 2021 were selected in the study, aged 44-94 years. And 100 healthy subjects were selected as healthy controls. According to estimated glomerular filtration rate (eGFR)and urine albumin-to-creatinine ratio (UACR), 200 subjects were divided into two subgroups: T2DM without DKD group (100 patients) and DKD group (100 patients). We investigated the differences of the characteristics, genotype frequency and allele distributions of rs112667 variants among different groups. RESULTS: The mean ages of the subjects with T2DM and healthy controls were 68.94 ± 7.18, 69.24 ± 8.9, respectively. The gender of subjects in the healthy controls were frequency-matched with the T2DM patients (50 males; 107 males, respectively). The TyG Index and the distribution of ALOX12 rs112667 genotype were significantly different between T2DM patients and healthy controls. The TyG Index and the AA genotype of rs112667 were associated with an increased risk of T2DM (P < 0.001, OR = 5.019; P = 0.024, OR = 2.451, respectively). The T2DM patients with the GG genotype of rs112667 had lower ACR lever compared with subjects with GA or AA genotype. The rs112667 genotype of (AA + GA) (P = 0.026, OR 2.419, 95% CI 1.112-5.266) was associated with an increased risk of DKD. CONCLUSION: The TyG Index is of important value for predicting T2DM.And ALOX12 gene polymorphism of rs112667 may play a role in pathological mechanism of the progression of diabetes in the Chinese population.
